Do you have case sensitive passwords ticked in wcConfig|System Security? 
PXTools password should be exactly the same as your WC login password. 
Maybe try changing your wc password and then try pxtools again. 
 
Irex v2.29 working fine here with Window 10 Pro 64bit 
look at the irex logs for errors 
are you running it in daemon mode?  rexw -daemon 
what do the semaphor files indicate?
