I did that for years.  Something like:
C DOS
D word processing and documents
E programs
F or I Windows
G games
H media
R S optical drives

At one point I had drive letters used up all the way through T.

We did that largely to reduce slack space due to cluster size inflation. 
NTFS made that unnecessary, so now it's

C Windows
D data
E more data
F G removables
H I J more data
N O external drives
R optical drive
